    The ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX) is a ready-to-use reagent with all the essential components for quantitative real-time PCR (qPCR) except primers and templates. The master mix contains a highly stable hot-start Taq polymerase in an optimized buffer with dsDNA specific SYBR green fluorescent dye. Consequently, the ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX) features high stability during storage (Fig. 1), even at 37°C for weeks. In addition to high sensitivity and signal intensity (Fig. 2), the ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX) performs a low background/ high specificity qPCR results (Fig. 3), as well as a better compatibility with fast PCR program (Fig. 4).
    With inert smart blue contrast dye, the ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX) is ready-to-use and greatly reduces pipetting errors, while largely improving the reproducibility of the process. The ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX) is also compatible with ROX reference dye if ROX is recommended by the manufacturer of the qPCR system. This master mix allows sensitive, precise amplification, real-time tracking of the amplification process, and simultaneous quantification for targeted DNA molecules.

    (A)Stability was analyzed via real-time PCR amplification with 1 ng cDNA as template, and TQ1200 ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX) stored at -20°C, 4°C, and 22°C for 30 days as reagents. The overlapped amplification and melt curves of Q-PCR reactions from differenet storage conditions display excellent stability of TQ1200 ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX).
    (B)TQ1200 ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX) and qPCR master mix from brand P and B were stored at -20°C and 37°C for 14 days for stability comparison. TQ1200 displays overlapped amplification curves between -20°C and 37°C groups showing highly consistent Ct value and fluorescent intensity. Inconsistent Ct value and decreased fluorescent intensity were observed in qPCR master mix from brand P and brand B, respectively.

    (A)The sensitivity assay of TQ1200 ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX) was examined with serial diluted cDNA from 25 ng to 1.4 fg as templates. The amplification curves display that TQ1200 is capable to amplify template as little as 1.4 fg, and over a wide range of template usage.

    The post-amplification melt-curve analysis was examined with 5 ng cDNA as template, relatively higher concentration of primers (400 nM), and TQ1200 ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX) or qPCR master mix from brand A as reagents. The single peak of melt curve displays a homogeneous PCR product amplified by TQ1200, while primer dimer signal was shown in qPCR results from brand A.

    The initial activation step of real-time PCR program was set as 2, 5, and 10 minutes at 95°C. TQ1200 ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX) contains a special hot-start Taq polymerase that omits time-consuming initial activation step required by chemically modified hot start Taq polymerases (Brand A). The overlapped amplification curves from different hot start duration display that TQ1200 ExcelTaq™ 2X Fast Q-PCR Master Mix (SYBR, no ROX) preforms successfully in short duration of initial activation (2 min).
